Are root negatively phototropic?
if yes, how to set up a experiment to test it?

Dear Fender,

It is negatively phototropic. But it is meaningless to know it because roots do not need light.
If you want to set up an experiment to test for it, then you can use set up as text book 3 which shows the figure of "Growth response of roots to gravity". Then add a light bulb to one of the side. There are more possible set ups. You can think about it, just follow the rule (i.e. one factor under study at each time and the other factors should not be unilateral.)
